It happened in Longden Road and saw a silver Audi upside down on its roof. The road reopened several hours later.
In a post on social media at 3.30pm on Wednesday the force told drivers to find alternative routes.
A follow up said officers expect the road to be closed for 'several hours' and that diversions were in place at the junctions with Belle Vue Gardens and South Hermitage
They said: "We are dealing with a two vehicle collision on Longden Road in Shrewsbury. The road is blocked, please find an alternative route."
Local residents have also reported seeing two air ambulances in the area, with one landing on the playing fields at Raby Crescent.
A spokesman for West Midlands Ambulance Service said: “We were called at 3.21pm to a road traffic collision on Longden Road in Shrewsbury. Two ambulances, a paramedic officer, the Midland Air Ambulance from Cosford and the Midland Air Ambulance from Tatenhill attended the scene.”
Shropshire Fire and Rescue Service is also at the scene and has sent two crews and an officer to the incident.
