The Chelsea loanee has not featured for Salop head coach Gareth Ainsworth since the heart-breaking last-gasp defeat to Charlton at The Valley almost two weeks ago.
Castledine is suffering from an 'overuse' injury with the boss saying Town must manage the 19-year-old's workloads accordingly.
And speaking in his press conference ahead of the trip to the New York Stadium this weekend, Ainsworth said the youngster will be struggling for the clash while saying there was positive news on the rest of the squad.
He said: "Leo's had this overuse-related injury, which happens when you're young.
"Taking them out is just as important as putting them in. These youngsters, it's really, really important for their development, mentally, physically, everything. So Leo's going to struggle for the squad on Saturday.
"Apart from him, everyone else is fit. Obviously, Toto has a match ban for his red card, but now everyone else is involved and ready to go.
"It's brilliant to see Morgan up and about again after his whack at Stockport, but I'm really looking forward to getting my teeth stuck into Rotherham."
