Wolves had the chance to go eight points clear of the relegation zone but goals from Ryan Sessegnon and Rodrigo Muniz condemned them to a 2-1 loss at Molineux.
With 12 games remaining, Wolves still have to play all three teams below them, with those results likely to decide whether they play Premier League football next season.
Wing-back Semedo told the club's website: "We knew that we were five points clear, but that's not enough. We have to keep going. We cannot slow down now.
"We have to carry on. We have to keep getting some points, especially when we play at home. We have a couple games now to come that are very important.
"Today was one of them. We couldn't win this game and now rest and think about the next game already."
Wolves were behind after 58 seconds, with Sessegnon left to run free on goal before coolly slotting home after Semedo was sucked into a challenge on Andreas Pereira .
Semedo held his hands up and took full responsibility for his side going a goal down.
He added: "I wasn't good enough with the decision I made, and that cost us a goal. We have to take the responsibility, and here I am for that.
"We have to work hard on that and try to come better. Usually, it's not a thing that happens to us, but today cannot happen again.
"We weren't focused, we weren't concentrated, and the beginnings of halves are very important."
Sessegnon was handed his first Premier League start of the season as Marco Silva made five changes from their 2-0 defeat to Crystal Palace at the weekend.
The former Tottenham full-back said: "I kind of knew it would be that kind of game. I'm ready and I'm happy I took the chance.
"I knew when I went one-on-one with the keeper, I'm very confident and very happy I scored.
"I had to make an impact. I love it when they (fans) sing my name, I feel very proud coming from the academy and this is my club.
"For the league it was important to get three points and bring this performance in for confidence for the FA Cup game."
